Metformin HCl
Indications/Uses
Maturity onset DM not responsive to diet alone.
Dosage/Direction for Use
Initially 500 mg tds or 850 mg bd. May gradually be increased up to max of 3 g/day.
Administration
Should be taken with food.
Contraindications
Hepatic or renal impairment. CV collapse, CHF, acute MI or other conditions leading to hypotension or hypoxaemia. DM complicated by acidosis, infection or gangrene during pregnancy or surgery. Pregnancy & lactation.
Special Precautions
Monitor for lactic acidosis. Determine serum creatinine levels prior to therapy & monitor regularly. Administration of iodinated contrast media. Discontinue therapy 48 hr prior to elective surgery w/ general anaesth. Co-administration w/ insulin & sulphonylureas. Childn 10-12 yr. Decreased renal function in elderly.
Adverse Reactions
Nausea, vomiting, diarrhoea, abdominal pain, loss of appetite, taste disturbance.
Drug Interactions
Vit B12, tetracycline, alcohol & alcohol-containing medications. Iodinated contrast agents. Glucocorticoids, β2-agonists, diuretics; ACE inhibitors.
